Cell Biology Laboratory 307 Schedule for Spring 2008
All linked materials must be studied PRIOR to your scheduled lab. You may wish to print the directions for the lab activities. Portions of the Worksheets must be completed PRIOR to the beginning of the lab. DO NOT print these labs all at once at the beginning of the semester. The instructor will be making final modifications to the labs up until the week of the lab exercise.
DateScheduled ActivityPrelab Worksheets Due
Jan 15/17Introduction to the course.

Lab exercises: Pipetting Exercise Worksheet, Phase Contrast Microscopy, Downloading, Image Capture and Microscopic Measurement, Microscopic Measurements Worksheet, Making Solutions Worksheet,

(Note: the Adipocyte Worksheet will not be used in 2008)
No prelab worksheets are due at the beginning of this lab. The Solutions Worksheet, the Pipetting Exercise Worksheet and the Microsopic Measurements Worksheets can be turned in for 5 points of credit each anytime prior to (but not after) the lab exam.
Jan 22/24Cell Fractionation and Determination of Protein Concentration; Instructions for using the Avanti Centrifuge; Nomogram; G-Force Table; Spectrophotometry; Generic Protein Assay (use for investigative lab)The PreLab portion of the Cell Fractionation Worksheet is due.
Jan 29/31Assay of SDH Activity
The Animation illustrating absorption and transmission may be useful in learning how a spectrophotometer works.
The PreLab portion of the SuccinateDH_Worksheet is due.
Feb 5/7Students Process Corn Leaves for Next Week's Lab
and then start their SDS-PAGE of Cellular Fractions. The Animation illustrating the principles of SDS-PAGE may be useful in helping students understand SDS-PAGE. All students must know how to graph with SemiLog paper. SemiLog paper (gif). The following two links do not need to be printed. They are provided to help you when you use Scion Image. Downloading Scion Image Using Scion Image. Access to the Semilog Spreadsheet
The PreLab portion of the SDS-PAGE_Worksheet. is due.
Feb 12/14Native Gel Electrophoresis of Peroxidases in Maize Leaves
The Animation illustrating the principles of Native (Nondenaturing) Gel Electrophoresis may be useful in helping to understand the principles of native gel electrophoresis.
The PreLab portion of the Native Gel Worksheet is due.
Feb 19/21Note: The following is a very long lab exercise. Thorough preparation is necessary to complete the lab in 3 hours. Come a little early if you can and start immediately when you arrive. Worked well as a one day lab in 2005.
Enzymes; Tyrosinase Activity; Standard Curves; Effect of Enzyme Concentration; Km; Vm
The PreLab portion of the Tyrosinase Worksheet is due.
Feb 26/28Isolation of Spinach Thylakoid Membranes and Measurement of Light Induced Electron Transport
These Emission Spectra may be useful for investigative labs later.
The PreLab portion of the Photosynthesis Worksheet is due.
Mar 1-9 Spring Break
Mar 11/13Review for Lab Exam Next Week Review sheet to aid in preparation for the lab exam. Students will be allowed to use a properly prepared (handwritten entries only except as indicated by the instructor) lab notebook and properly completed lab worksheets to take the lab exam.
Mar 18/20Lab Exam
Mar 25/27 Begin Planning for the Investigative Lab. Establish groups of 2 or 3 students. Read the course syllabus for specific limitations on the spring 2008 investigative lab. Print out a copy of the "Investigative Lab Proposal" and read over the questions to get an idea of what you will be doing. DO NOT FILL IT OUT! Come to lab and discuss your investigative lab ideas with the instructor. The instructor will have insights relating to whether or not your proposed experiments are feasible with available equipment, time and supplies. Once the instructor has verbally approved your proposal, work as a group to thoroughly complete one "Investigative Lab Proposal" and hand it in before leaving the lab.
Apr 1/3Begin Experiments
Apr 8/10 Continue Experiments
Apr 15/17Continue Experiments
Apr 22/24Oral Presentations, Hand in Written Project Report, Each student must bring one instructor evaluation form to lab. The student's name and the title of the presentation should be typed on the evaluation form. Each student should also bring enough copies of the Peer Evaluation Form for each of the other students in the class.
